What does substantial improvement mean for an existing building?

Generally you must more than double the allocable building basis over a 30-month period after acquisition (with exclusions for land and certain unimproved scenarios). Mixed-use and phased construction need a line-item basis allocation — not a back-of-napkin SF cost.
